A mutation in DNA can affect a protein being made because the bases in DNA will either be changed, added to, or taken away as a result of the mutation. This will lead to a different base sequence, leading to a different amino acid being coded. The order of amino acids in a strand is specific to a protein. Just one change in an amino acid will change the protein.
