As an intern in a biology institute, I have a 17.3kbp plasmid that needs to be transformed into E. coli. However, I have tried multiple times and have had no or very few colonies on the LB plate. I am aware that large plasmids have a lower frequency of transformation. How can I improve the chances of successful transformation?
